Accurate measurements of myelin volume in the brain are of great interest for both medical research and clinical evaluations. Such measurements can provide an excellent diagnostic information for detecting neurological diseases in early stages, and it would potentially also allow for effective treatment of a neurological disease. Moreover it can be used as a research tool for researchers when trying to understand brain function as well as for disease prevention. However, existing methods for measuring myelin volume may not describe the exchange of water in-between the myelin sheets and cellular water entirely accurately. In this thesis, a set of models were both developed and implemented in order to investigate if it was possible to determine the amount of myelin and the exchange between pools in the brain based on data that was obtained using magnetic resonance spectroscopy (mrs). Ten healthy controls were examined using mrs with a focus on water exchange within brain tissue. A two-pool model that include water exchange and a two-pool model without such exchange were both useful models for describing the experimental data. Moreover, it was also possible to determine the total volume of myelin using the two-pool model without water exchange. In contrast, it was not possible to determine myelin volume sufficiently accurate using the two-pool model that included water exchange, although it was possible to determine the water exchange rate in such a system.
