    Abstract [en]

This study is intended to investigate the prerequisites for the principals to perform their work and whether they differ between schools. This has been investigated by studying the underlying management structure and the management instrument and how they have shaped the prerequisites for the principals. The study is based on interviews with princi- pals and a quality director from both public school and private school. The study shows that the management instruments goals, communication, mandate and decision as well as support functions, and to some extent the management structure, shapes the prerequisites for the principals. These management instrument have also been looked into more thor- oughly, in order to see how well they work to create the right conditions for the principals in the public school and private school. From the results that was obtained from this study, we could see that the management instruments don’t work optimally in order to give the right prerequisites for the principals. Mainly have the results shown that the principals in the private school have better perquisites than the principals working in the public school.

Ett försprång till skapande - en undersökning om förutsättningarna för Musikproduktion på Kulturskolan2008Independent thesis Basic level (degree of Bachelor), 10 credits / 15 HE creditsStudent thesis
    Abstract [sv]

Utbildning, kompetens och arbete2002Collection (editor) (Other academic)
    Abstract [sv]

Användning av prototyper som verktyg för kravhantering i agil mjukvaruutveckling: - En fallstudie2018Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

Requirements Engineering (RE) in Agile Software Development (ASD) is a challenge thatmany face and several techniques exist when doing so. One such technique is prototyping, when a model of a product is used to gather important information in software develop-ment. To describe how much a prototype resembles the product the notion of fidelity is used. The aim of this study is to contribute to research regarding prototyping in ASD,and to examine the effect of a prototype’s fidelity when using prototypes in discussionsduring RE. A case study is performed at the company Exsitec where staff are interviewedregarding prototyping in software development. Thereafter, two prototypes of low andhigh fidelity are developed and used in interviews as a basis for discussion. Based on thisstudy, the use of prototypes in software projects can help customers trust the process, improve communication with customers, and facilitate when trying to reach consensusamong different stakeholders. Furthermore, depending on how they are used, prototypescan contribute to understanding the big picture of the requirements and can also serve asdocumentation. The study also shows some, albeit subtle, differences in the informationcollected using prototypes with low and high fidelity. The use of a high fidelity prototypeseems to generate more requirements, but makes interviewees less likely to come up withlarger, more comprehensive requirement changes.

Fristående skolor1999Independent thesis Basic level (professional degree)Student thesis
    Abstract [sv]

Improving methane production using hydrodynamic cavitation as pre-treatment2016Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

To develop anaerobic digestion (AD), innovative solutions to increase methane yields in existing AD processes are needed. In particular, the adoption of low energy pre-treatments to enhance biomass biodegradability is needed to provide efficient digestion processes increasing profitability. To obtain these features, hydrodynamic cavitation has been evaluated as an innovative solutions for AD of waste activated sludge (WAS), food waste (FW), macro algae and grass, in comparison with steam explosion (high energy pre-treatment). The effect of these two pre-treatments on the substrates, e.g. particle size distribution, soluble chemical oxygen demand (sCOD), biochemical methane potential (BMP) and biodegradability rate, have been evaluated. After two minutes of hydrodynamic cavitation (8 bar), the mean fine particle size decreased from 489- 1344 nm to 277- 381 nm (≤77% reduction) depending of the biomasses. Similar impacts were observed after ten minutes of steam explosion (210 °C, 30 bar) with a reduction in particle size between 40% and 70% for all the substrates treated.  In terms of BMP value, hydrodynamic cavitation caused significant increment only within the A. nodosum showing a post treatment increment of 44% compared to the untreated value, while similar values were obtained before and after treatment within the other tested substrates. In contrast, steam explosion allowed an increment for all treated samples, A. nodosum (+86%), grass (14%) and S. latissima (4%). However, greater impacts where observed with hydrodynamic cavitation than steam explosion when comparing the kinetic constant K. Overall, hydrodynamic cavitation appeared an efficient pre-treatment for AD capable to compete with the traditional steam explosion in terms om kinetics and providing a more efficient energy balance (+14%) as well as methane yield for A. nodosum.

A platform for third-party applications on the web2013Independent thesis Basic level (degree of Bachelor), 10,5 credits / 16 HE creditsStudent thesis
    Abstract [en]

A trend seen on the web today is to create a platform where externally developed applications can run inside some kind of main application. This is often done by providing an API to access data and business logic of your service and a sandbox environment in which third-party applications can run. By providing this, it is made possible for external developers to come up with new ideas based on your service. Some good examples on this are Spotify Apps, Apps on Facebook and SalesForce.com.

Ipendo Systems AB is a company that develops a web platform for intellectual properties. Currently most things on this platform are developed by developers at Ipendo Systems AB. Some interest has though risen to enable external developers to create applications that will in some way run inside the main platform.

In this thesis an analysis of already existing solutions has been done. These solutions were Spotify Apps and Apps on Facebook. The two have different approaches on how to enable third-party applications to run inside their own service. Facebook’s solution builds mainly on iframe embedded web pages where data access is provided through a web API. Spotify on the other hand hosts the third-party applications themselves but the applications may only consist of HTML5, CSS3 and JavaScript.

In addition to the analysis a prototype was developed. The purpose of the prototype was to show possible ways to enable third-party applications to run inside your own service. Two solutions showing this were developed. The first one was based on Facebook’s approach with iframing of external web pages. The second was a slightly modified version of Spotify’s solution with only client-side code hosted by the main application. To safely embed the client side code in the main application a sandboxing tool for JavaScript called Caja was used.

Of the two versions implemented in the prototype was the Iframe solution considered more ready to be utilized in a production environment than Caja. Caja could be seen as an interesting technique for the future but might not be ready to use today. The reason behind this conclusion was that Caja decreased the performance of the written JavaScript as well as adding complexity while developing the third-party applications.

    Abstract [sv]

    Abstract [sv]

    Abstract [en]

"Northern Lights in Logistics & Supply Chain Management" portrays the past, present and future research of the subject in the Nordic countries. The NOFOMA conference - a network of Nordic researchers within the field of Logistics and Supply Chain Management - has been a focal point in the contribution to the continuous improvement and further development of Nordic research. The network has also opened up for interaction with fellow researchers from other countries. There are sixteen chapters in the book that in its own way colours the Nordic rainbow of research within Logistics and Supply Chain Management. The chapters are structured in four themes: 1) Origins and strategic aspects; 2) Research approaches in the Nordic countries; 3) Advancement of distribution strategies and; 4) Emerging application areas of logistics and SCM. The chapters provide an understanding and, perhaps more importantly, consciousness for scholars that are part of this research environment: Where are we now, what have we been influenced by, and in what area are we able to provide positive impact? The aim of the book is also to contribute to increased visibility to fellow international scholars within Logistics and Supply Chain Management.

    Abstract [en]

Based on different observations, in theory as well as in practice, we have identified strategies and operations following two different tracks, striving in different directions, despite it being well known they should go hand in hand. For companies challenged by a more and more dynamic business environment with heavier market segmentation, additional marketing channels, increasing globalisation on supplier and customer side, and high pressure on profit margins, the result from this is competitive weakness. The purpose of this article is to switch focus from operational effectiveness and strategic positioning as static success concepts on how to improve industrial distribution, to dynamic challenges of how to continuously manage the interaction between marketing strategies and operations (e.g., logistics) under the influence of a dynamic business environment. From theories in market strategies, logistics, flexibility, and marketing channels together with empirical experience from a best practice case study, we are introducing a model for dynamic effectiveness, describing the different characteristics of a company and what to focus on in order to become more dynamic. In order to constantly move to new market positions, and at the same time, restructure logistics and improve operational effectiveness, we have identified the dynamic capabilities of an organisation to be the key to success in industrial distribution. We define dynamic effectiveness as "how fast-and-well a company can go from one strategic positioning and productivity frontier to another." It tells that a frequent interaction between new strategic moves and actions for higher operational effectiveness is required to be in pace with the dynamic and changing business environment and to stay ahead of competition. As a part, of this, best, practice logistics performance makes it possible not only to be more agile to new strategic moves, but also to drive strategic development from a high operational level. This is achieved by designing logistics to be a resource base to support and be an enabler for new strategic moves on the market. © 2004 by The Haworth Press, Inc. All rights reserved.

Mastering Supply Chain Management in an era of uncertainty at SKF2015In: Global Business and Organizational Excellence: a review of research & best practices, ISSN 1932-2054, Vol. 34, no 6, p. 6-17Article in journal (Other academic)
    Abstract [en]

To stay ahead in an increasingly competitive business environment, organizations need agile supply chain systems that are holistically designed and managed. The experiences of SKF, a Swedish multinational firm, point to the benefits of reengineering upstream capabilities to create value downstream. To foster the flexibility, responsiveness, and other dynamic capabilities needed to manage increased supply chain complexity, the company established programs to involve its suppliers in the value creation process, making them an extension of the organization’s resource base. The overall objective has been to migrate from a business model based on economies of scale in operations to one that exploits both economies of scope and economies of integration. As a result of its efforts to create a more agile and cost-efficient supply chain, the company has been able to decrease supply risks while improving its value creation process and responsiveness to new customer demands and advancing its sustainability and social responsibility initiatives.

    Abstract [sv]

Inside the Core: Managing risks in Business Critical Outsourcing2011In: Proceedings of the 20th Annual IPSERA Conference, Vision 20/20 – Preparing Today for Tomorrow’s Challenges / [ed] F. Rozemeijer, M. Wetzels, L. Quintens, 2011, p. 1092-1101Conference paper (Refereed)
    Abstract [en]

Organizations are increasingly outsourcing business critical activities, which increases the risk of substantial negative impact on the financial and strategic business performance. The aim of this paper is to show how outsourcer and supplier seek to mitigate the risks during the negotiations and the ongoing operations of the outsourced activities. The conclusion is that risk mitigating dimensions are directional and operates at different organizational levels, and secondly, that the dimension operates sequentially as a means to mitigate performance and relational risks. It is also concluded that goodwill trust and a cooperative stance have substantial impact on the success of the outsourcing.

Managing risk in complex and business critical outsourcing of services2011In: Proceedings of 18th International Annual Conference of the European Operations Management Association. EurOMA, 2011Conference paper (Refereed)
    Abstract [en]

Over the past two decades the phenomenon of outsourcing of resources and activities has emerged as an important trend in a wide range of organizations and many companies are today outsourcing complex and business critical resources and activities. This increase in business criticality also increases the risk of substantial and negative impact on the financial and strategic performance. The proposition is that business critical outsourcing is approached with a joint risk perspective and that an understanding of the decision process is essential for success. Further, it is important to view outsourcing as an ongoing process with a number of key stages.

The Role of Logistics in Retailers' Corporate Strategy: A Driver for Growth and Customer Value2010In: Supply Chain Forum: an International Journal, ISSN 1625-8312, E-ISSN 1624-6039, Vol. 11, no 4, p. 14-23Article in journal (Refereed)
    Abstract [en]

This article explores the role of logistics and supply chain management (SCM) in retailers’ corporate strategy and is based on reviewing multiple qualitative case studies of companies in the Swedish food retail sector. The article proposes that the role of logistics for what is referred to in this paper as modern retailers is twofold: to create profitability and to support growth and market expansion. International modern retailers are empirically compared with similar companies in the Swedish market. The dominating Swedish food retailers have taken command in the food supply chain but are primarily concerned with traditional logistics roles in cost cutting. This role is perfectly fine as long as the strategic intent of the companies is focused on market retention and not on geographical expansion and/or new marketing or store concepts. We argue that the success of modern retailers in terms of growth rate, profitability, and market expansion is explained to a large extent by the role of logistics in corporate strategy—when the role for supporting growth and increasing customer value from high-performance supply capabilities is a driver for market expansion. However, in many of our cases the traditional logistics roles of cutting costs and lead-times is still the dominating pattern that limits the possibility of expanding into new markets. Better knowledge of logistics’ role in retail corporate strategy can help companies develop and improve their strategic intent as well as redefine the role of logistics.

Logistik i svensk handel: Ett projekt finansierat av Handelns Utvecklingsråd2011Report (Other academic)
    Abstract [sv]

Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [sv]

A Gröbner basis algorithm for fast encoding of Reed-Müller codes2016Independent thesis Basic level (degree of Bachelor), 10,5 credits / 16 HE creditsStudent thesis
    Abstract [en]

In this thesis the relationship between Gröbner bases and algebraic coding theory is investigated, and especially applications towards linear codes, with Reed-Müller codes as an illustrative example. We prove that each linear code can be described as a binomial ideal of a polynomial ring, and that a systematic encoding algorithm for such codes is given by the remainder of the information word computed with respect to the reduced Gröbner basis. Finally we show how to apply the representation of a code by its corresponding polynomial ring ideal to construct a class of codes containing the so called primitive Reed-Müller codes, with a few examples of this result.

Hide and Seek in a Social Network2017Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

In this thesis a known heuristic for decreasing a node's centrality scores while maintaining influence, called ROAM, is compared to a modified version specifically designed to decrease eigenvector centrality. The performances of these heuristics are also tested against the Shapley values of a cooperative game played over the considered network, where the game is such that influential nodes receive higher Shapley values. The modified heuristic performed at least as good as the original ROAM, and in some instances even better (especially when the terrorist network behind the World Trade Center attacks was considered). Both heuristics increased the influence score for a given targeted node when applied consecutively on the WTC network, and consequently the Shapley values increased as well. Therefore the Shapley value of the game considered in this thesis seems to be well suited for discovering individuals that are assumed to actively trying to evade social network analysis.

Mer lättläst: Påbyggnad av ett automatiskt omskrivningsverktyg till lätt svenska2011Independent thesis Basic level (degree of Bachelor), 12 credits / 18 HE creditsStudent thesis
    Abstract [sv]

Combined Platform for Boost Guidance and Attitude Control for Sounding Rockets2004Independent thesis Basic level (professional degree)Student thesis
    Abstract [en]

This report handles the preliminary design of a control system that includes both attitude control and boost control functionality for sounding rockets. This is done to reduce the weight and volume for the control system.

A sounding rocket is a small rocket compared to a satellite launcher. It is used to launch payloads into suborbital trajectories. The payload consists of scientific experiments, for example micro-gravity experiments and astronomic observations. The boost guidance system controls the sounding rocket during the launch phase. This is done to minimize the impact dispersion. The attitude control system controls the payload during the experiment phase.

The system that is developed in this report is based on the DS19 boost guidance system from Saab Ericsson Space AB. The new system is designed by extending DS19 with software and hardware. The new system is therefore named DS19+. Hardware wise a study of the mechanical and electrical interfaces and also of the system budgets for gas, mass and power for the system are done to determine the feasibility for the combined system.

Further a preliminary design of the control software is done. The design has been implemented as pseudo code in MATLAB for testing and simulations. A simulation model for the sounding rocket andits surroundings during the experiment phase has also been designed and implemented in MATLAB. The tests and simulations that have been performed show that the code is suitable for implementation in the real system.

Comparing modifiability of React Native and two native codebases2017Independent thesis Advanced level (degree of Master (Two Years)), 20 credits / 30 HE creditsStudent thesis
    Abstract [en]

Creating native mobile application on multiple platforms generate a lot of duplicate code. This thesis has evaluated if the code quality attribute modifiability improves when migrating to React Native. One Android and one iOS codebase existed for an application and a third codebase was developed with React Native. The measurements of the codebases were based on the SQMMA-model. The metrics for the model were collected with static analyzers created specifically for this project. The results created consists of graphs that show the modifiability for some specific components over time and graphs that show the stability of the platforms. These graphs show that when measuring code metrics on applications over time it is better to do this on a large codebase that has been developed for some time. When calculating a modifiability value the sum of the metrics and the average value of the metrics between files should be used and it is shown that the React Native platform seems to be more stable than native.

Maternal heterozygosity and progeny fitness association in an inbred Scots pine population2013In: Genetica, ISSN 0016-6707, E-ISSN 1573-6857, Vol. 141, no 1-3, p. 41-50Article in journal (Refereed)
    Abstract [en]

Associations between heterozygosity and fitness traits have typically been investigated in populations characterized by low levels of inbreeding. We investigated the associations between standardized multilocus heterozygosity (stMLH) in mother trees (obtained from12 nuclear microsatellite markers) and five fitness traits measured in progenies from an inbred Scots pine population. The traits studied were proportion of sound seed, mean seed weight, germination rate, mean family height of one-year old seedlings under greenhouse conditions (GH) and mean family height of three-year old seedlings under field conditions (FH). The relatively high average inbreeding coefficient (F) in the population under study corresponds to a mixture of trees with different levels of co-ancestry, potentially resulting from a recent bottleneck. We used both frequentist and Bayesian methods of polynomial regression to investigate the presence of linear and non-linear relations between stMLH and each of the fitness traits. No significant associations were found for any of the traits except for GH, which displayed negative linear effect with stMLH. Negative HFC for GH could potentially be explained by the effect of heterosis caused by mating of two inbred mother trees (Lippman and Zamir 2006), or outbreeding depression at the most heterozygote trees and its negative impact on the fitness of the progeny, while their simultaneous action is also possible (Lynch. 1991). However, since this effect wasn’t detected for FH, we cannot either rule out that the greenhouse conditions introduce artificial effects that disappear under more realistic field conditions.

Can Lactobacillus Reuteri Prevent Allergic Disease in Early Childhood?2009Doctoral thesis, comprehensive summary (Other academic)
    Abstract [en]

Background: An altered microbial exposure may be partly responsible for the increase of allergic diseases in populations with a western lifestyle. Activation of the immune system by microbes early in life is probably required for an accurate maturation of the immune system. Probiotics, live bacteria which are considered to confer health when ingested, have been suggested to prevent eczema and sensitisation infants.

Aim: The general aim of this thesis was to assess the effect of oral supplementation with the probiotic bacterium Lactobacillus reuteri (L. reuteri) in infancy on the development of allergic disease and sensitisation during the first 2 years of life and to examine mechanisms possibly underlying eventual effects on allergic manifestations.

Subjects: The thesis is based on results obtained from a prospective double-blind placebo-controlled multicenter trial, comprising 232 families with allergic disease, of whom 188 completed the study.

Methods: The families were recruited at the antenatal clinic, and the mothers received L. reuteri ATCC 55730 (1 x 108 colony forming units) or placebo daily from gestational week 36 until delivery. Their babies then continued with the same study product from birth until 12 months of age and were followed up for another year. The primary outcomes were allergic disease, with or without positive skin prick test or circulating IgE to food allergens. Bacterial counts and prevalence were assessed in maternal breast milk and faeces and infant faeces, employing conventional cultivation methods. Cytokines and IgA antibodies were analysed in colostrum and mature milk from the mothers with ELISA, and Na/K- ratio in breast milk with ion selective electrodes. Circulating Th1/Th2-associated chemokines were analysed in cord and peripheral blood in the infants with Luminex or ELISA technique.

Results: The incidence of eczema was similar, 36% in the treated versus 34% in the placebo group. The L. reuteri group had a lower cumulative incidence of IgE-associated allergic disease, 20% versus 35% (p=0.04), and less IgE-associated eczema during the second year, 8% versus 20% (p=0.02). The prevalence of L. reuteri was higher during the first year of life in stool samples from infants, as well as in colostrum, in the active as compared to the placebo treated group. Colostrum from L. reuteri supplemented mothers had lower levels of TGF-β2, and low levels of this cytokine were associated with less sensitisation. Low Th1- and high Th2-associated chemokine levels preceded allergic disease. The presence of L. reuteri in stool was associated with lower levels of the Th2-associated chemokines CCL17 and CCL22 and higher levels of the Th1-associated CXCL11.

Conclusion: Although a preventive effect of probiotics on infant eczema was not confirmed, the L. reuteri treated infants had lower incidence of IgE-associated allergic disease at two years of age, and therefore possibly run a reduced risk to develop later respiratory allergic disease. The mechanisms underlying this effect require further elucidation.

    Abstract [en]

BACKGROUND: An altered microbial exposure may underlie the increase of allergic diseases in affluent societies. Probiotics may alleviate and even prevent eczema in infants.

OBJECTIVE: To prevent eczema and sensitization in infants with a family history of allergic disease by oral supplementation with the probiotic Lactobacillus reuteri.

METHODS: Double-blind, randomized, placebo-controlled trial, which comprised 232 families with allergic disease, of whom 188 completed the study. The mothers received L reuteri ATCC 55730 (1 x 10(8) colony forming units) daily from gestational week 36 until delivery. Their babies then continued with the same product from birth until 12 months of age and were followed up for another year. Primary outcome was allergic disease, with or without positive skin prick test or circulating IgE to food allergens.

RESULTS: The cumulative incidence of eczema was similar, 36% in the treated versus 34% in the placebo group. The L reuteri group had less IgE-associated eczema during the second year, 8% versus 20% (P = .02), however. Skin prick test reactivity was also less common in the treated than in the placebo group, significantly so for infants with mothers with allergies, 14% versus 31% (P = .02). Wheeze and other potentially allergic diseases were not affected.

CONCLUSION: Although a preventive effect of probiotics on infant eczema was not confirmed, the treated infants had less IgE-associated eczema at 2 years of age and therefore possibly run a reduced risk to develop later respiratory allergic disease. CLINICAL IMPLICATION: Probiotics may reduce the incidence of IgE-associated eczema in infancy.

    Abstract [en]

OBJECTIVES: This is to identify factors affecting the prevalence of Lactobacillus reuteri in maternal faeces and breast milk and infant faeces after oral supplementation with L reuteri and to assess the influence on microbial ecology, particularly Clostridium difficile and Bifidobacterium colonization.

MATERIALS AND METHODS: In this double-blind trial, 232 mothers with a family history of atopic disease were randomized to a daily intake of either L reuteri American-type culture collection (ATCC) 55730 (1 x 10 colony-forming units [CFU]) or placebo for the last 4 weeks of pregnancy. Their babies then continued with the same study product daily from birth until 12 months of age. Bacterial counts and prevalence were assessed in maternal breast milk and faeces and infant faeces, using conventional cultivation methods.

RESULTS: The prevalence of L reuteri was higher during the first year of life in the stool samples from infants in the active as compared with the placebo-treated group. The highest prevalence was recorded at 5 to 6 days of age (82% in the treated vs 20% in the placebo group, P < 0.001). Lactobacillus reuteri was isolated from 12% and 2%, respectively, in the colostrum samples (P < 0.05). Breast-feeding seemed to reduce faecal L reuteri counts, although antibiotics did not influence the levels of L reuteri. The administration of L reuteri did not affect bifidobacteria or C difficile colonization.

CONCLUSION: Lactobacillus reuteri may be detected in breast milk after oral supplementation to the mother and in almost all infants after oral supplementation during the first year of life, as well as occasionally in many untreated infants.

    Abstract [en]

The immunological composition of breast milk differs between mothers. The reasons for these differences and the consequences for the breast-fed infants are poorly understood. The aim of this study was to evaluate the effect of probiotic Lactobacillus reuteri supplementation on the immunological composition of breast milk in relation to sensitization and eczema in the babies. Total IgA, secretory IgA (SIgA), TGF-beta1, TGF-beta2, IL-10, TNF, soluble CD14 (sCD14), and Na/K ratios were analyzed in colostrum and mature milk obtained from women treated with L. reuteri (n = 54) or placebo (n = 55) from gestational week 36 until delivery. Bacteriological analyses of L. reuteri were performed in faecal samples of the mothers. The infants were followed prospectively for 2 yr regarding development of eczema and sensitization as defined by a positive skin prick test and/or circulating allergen-specific IgE antibodies at 6, 12, and 24 months of age. Supplementation of L. reuteri during pregnancy was associated with low levels of TGF-beta2 and slightly increased levels of IL-10 in colostrum. For TGF-beta2, this association was most pronounced in mothers with detectable L. reuteri in faeces. Infants receiving breast milk with low levels of TGF-beta2 were less likely to become sensitized during their first 2 yr of life. A similar trend was observed for development of IgE-associated eczema. The levels of total IgA, SIgA, TGF-beta1, TNF, sCD14, and Na/K ratios in breast milk were not affected by the intake of L. reuteri. None of these parameters correlated with sensitization or development of eczema in the infant, except for high Na/K ratios that associated with increased risk of sensitization. Supplementation with L. reuteri during late pregnancy reduces breast milk levels of TGF-beta2, and low levels of this cytokine are associated with less sensitization and possibly less IgE-associated eczema in breast-fed infants.

    Abstract [en]

Background: Analyses of circulating chemokines offer novel tools to investigate the Th1/Th2 imbalance in allergic disease in vivo and explore the influence of pre- and postnatal factors in infancy.

Objective: To relate circulating Th1- and Th2-associated chemokines to the development of allergic disease, pre- and postnatal factors and probiotic supplementation in infancy.

Methods: Circulating levels of Th1-associated CXC-chemokine ligand (CXCL)9, CXCL10 and CXCL11 and Th2-associated CC-chemokine ligand (CCL)17, CCL18 and CCL22 were assessed with Luminex and ELISA at birth (n=109), 6 (n=104), 12 (n=116) and 24 months (n=123) in 179 infants completing a double-blind placebo-controlled allergy prevention trial with Lactobacillus reuteri during the last month of gestation and through the first year of life. The infants were followed regarding development of allergic disease and sensitization until two years of age.

Results: The Th2-associated chemokines were as highest at birth and then decreased, whereas the Th1-associated chemokines increased with age. Low Th1- and high Th2-associated chemokine levels were observed in children developing allergic disease. Sensitization was preceded by elevated CCL22 and reduced CXCL11 levels. High Th2-associated chemokine46 levels were associated with increased birth length and weight and long duration of breastfeeding, and high Th1-associated chemokine levels with day-care attendance. Presence of L. reuteri in stool the first week of life was associated with low CCL17 and CCL22 and high CXCL11 levels at 6 months.

Conclusion: Allergic disease in infancy was associated with low circulating Th1- and high Th2-associated chemokine levels during the first year of life. The chemokine levels were affected by both pre and –postnatal factors.

A Finite Element Model Updating Formulation Using Frequency Responses and Eigenfrequencies1996Report (Other academic)
    Abstract [en]

A novel frequency and modal domain formulation of the model updating problem is presented. Deviations in discrete frequency responses and eigenfrequencies, between the model to be updated and a reference model, constitute the criterion function. A successful updating thus results in a model with the reference's input-output relations at selected fre- quencies. The formulation is demonstrated to produce a criterion function with a global minimum having a large domain of attraction with respect to stiffness and mass variations. The method relies on mode grouping and uses a new extended modal assurance criterion number (eMAC) for identifying related modes. A quadratic objective with inexpensive evaluation of approximate Hessians give a rapid convergence to a minimum by the use of a regularized Gauss-Newton method. Physical bounds on parameters and complementary data, such as structural weight, are treated by imposing set constraints and linear equality constraints. Efficient function computation is obtained by model reduction using a moderately sized base of modes which is recomputed during the minimization. Statistical properties of updated parameters are discussed. A verification example show the performance of the method.

Low gut microbiota diversity in early infancy precedes asthma at school age2014In: Clinical and Experimental Allergy, ISSN 0954-7894, E-ISSN 1365-2222, Vol. 44, no 6, p. 842-850Article in journal (Refereed)
    Abstract [en]

    BACKGROUND:

Low total diversity of the gut microbiota during the first year of life is associated with allergic diseases in infancy, but little is known how early microbial diversity is related to allergic disease later in school age.

    OBJECTIVE:

To assess microbial diversity and characterize the dominant bacteria in stool during the first year of life in relation to the prevalence of different allergic diseases in school age, such as asthma, allergic rhinoconjunctivitis (ARC) and eczema.

    METHODS:

The microbial diversity and composition was analysed with barcoded 16S rDNA 454 pyrosequencing in stool samples at 1 week, 1 month and 12 months of age in 47 infants which were subsequently assessed for allergic disease and skin prick test reactivity at 7 years of age (ClinicalTrials.gov ID NCT01285830).

    RESULTS:

Children developing asthma (n = 8) had a lower diversity of the total microbiota than non-asthmatic children at 1 week (P = 0.04) and 1 month (P = 0.003) of age, whereas allergic rhinoconjunctivitis (n = 13), eczema (n = 12) and positive skin prick reactivity (n = 14) at 7 years of age did not associate with the gut microbiota diversity. Neither was asthma associated with the microbiota composition later in infancy (at 12 months). Children having IgE-associated eczema in infancy and subsequently developing asthma had lower microbial diversity than those that did not. There were no significant differences, however, in relative abundance of bacterial phyla and genera between children with or without allergic disease.

    CONCLUSION AND CLINICAL RELEVANCE:

Low total diversity of the gut microbiota during the first month of life was associated with asthma but not ARC in children at 7 years of age. Measures affecting microbial colonization of the infant during the first month of life may impact asthma development in childhood.
